Cadmium (Cd) accumulations in a Cd hyper‐accumulator fern, Athyrium yokoscense (Ay), and tobacco, Nicotiana tabacum (Nt), were kinetically analysed using the positron‐emitting tracer imaging system under two medium conditions (basal and no‐nutrient). In Ay, maximumly 50% and 15% of the total Cd accumulated in the distal roots and the shoots under the basal condition, respectively. Interestingly, a portion of the Cd in the distal roots returned to the medium. In comparison with Ay, a little fewer Cd accumulations in the distal roots and clearly higher Cd migration to the shoots were observed in Nt under the basal condition (maximumly 40% and 70% of the total Cd, respectively). The no‐nutrient condition down‐regulated the Cd migration in both species, although the regulation was highly stricter in Ay than in Nt (almost no migration in Ay and around 20% migration in Nt). In addition, the present work enabled to estimate physical and physiological Cd accumulation capacities in the distal roots, and demonstrated condition‐dependent changes especially in Ay. These results clearly suggested occurrences of species‐/condition‐specific regulations in each observed parts. It is probable that integration of these properties govern the specific Cd tolerance/accumulation in Ay and Nt.  相似文献

1. Factors affecting host preference in herbivorous insects are actively discussed. Larval performance, competition and predation on each host, and host abundance are reportedly factors affecting the host preference of ovipositing females. In the present study, chronological changes in female host preference were examined by employing a flea weevil species that uses the original host and a newly introduced host simultaneously. 2. Orchestes hustachei Klima (Coleoptera: Curclionidae) is an aphidophagous flea weevil that oviposits on aphid galls, and the larvae feed on aphids inside. The weevil's native hosts are galls of the Tetraneura species on Ulmus davidiana, while the novel hosts are galls of an introduced species, Paracolopha morrisoni on Zelkova serrata, an introduced hardwood. Choice experiments were conducted using Tetraneura galls and Paracolopha galls and the results were compared with experiments conducted 10 years ago. 3. More than 90% of ovipositing females selected Paracolopha galls. This result is in marked contrast with the result of the 2002 experiment, in which 66.3% of females selected Paracolopha galls. To explore driving forces of the preference change, mortality factors, pupal mass on the two hosts, and temporal changes in the abundance of the host galls were examined. 4. Abundance of Tetraneura galls decreased gradually throughout the last 26 years. By contrast, weevil survival and performance did not vary significantly between the two hosts. Therefore, it is concluded that temporal changes in the relative abundance of two hosts are main factors of the change in host preference.  相似文献

The translational GTPases promote initiation, elongation, and termination of protein synthesis by interacting with the ribosome. Mutations that impair GTP hydrolysis by eukaryotic translation initiation factor 5B/initiation factor 2 (eIF5B/IF2) impair yeast cell growth due to failure to dissociate from the ribosome following subunit joining. A mutation in helix h5 of the 18S rRNA in the 40S ribosomal subunit and intragenic mutations in domain II of eIF5B suppress the toxic effects associated with expression of the eIF5B-H480I GTPase-deficient mutant in yeast by lowering the ribosome binding affinity of eIF5B. Hydroxyl radical mapping experiments reveal that the domain II suppressors interface with the body of the 40S subunit in the vicinity of helix h5. As the helix h5 mutation also impairs elongation factor function, the rRNA and eIF5B suppressor mutations provide in vivo evidence supporting a functionally important docking of domain II of the translational GTPases on the body of the small ribosomal subunit.  相似文献

Photosynthetic nitrite reductase (PNiR) was purified extensivelyfrom spinach leaves by the method including acetone precipitation,ammonium sulfate fractionations and DEAE-cellulose column chromatography.The purified PNiR had the highest specific activity thus farobtained. Absorption spectrum of the purified PNiR indicatedthat it had no flavin component. There was no difference inthe rate of photosynthetic nitrite reduction between aerobicand anaerobic conditions. In chloroplasts two reductase systems related with ferredoxinhad been recognized, which were ferredoxin-nitrite and ferredoxin-NADPreductase system. The relationship between the two systems wasinvestigated by using the reconstituted enzyme system. Whenthe reduction of NADP was taking place, the reduction of nitritewas strongly inhibited. Eukaryotic initiation factor 1A (eIF1A) and the GTPase IF2/eIF5B are the only universally conserved translation initiation factors. Recent structural, biochemical and genetic data indicate that these two factors form an evolutionarily conserved structural and functional unit in translation initiation. Based on insights gathered from studies of the translation elongation factor GTPases, we propose that these factors occupy the aminoacyl-tRNA site (A site) on the ribosome, and promote initiator tRNA binding and ribosomal subunit joining. These processes yield a translationally competent ribosome with Met-tRNA in the ribosomal peptidyl-tRNA site (P site), base-paired to the AUG start codon of a mRNA.  相似文献

Abstract

In addition to the small and large ribosomal subunits, aminoacyl-tRNAs, and an mRNA, cellular protein synthesis is dependent on translation factors. The eukaryotic translation initiation factor 5A (eIF5A) and its bacterial ortholog elongation factor P (EF-P) were initially characterized based on their ability to stimulate methionyl-puromycin (Met-Pmn) synthesis, a model assay for protein synthesis; however, the function of these factors in cellular protein synthesis has been difficult to resolve. Interestingly, a conserved lysine residue in eIF5A is post-translationally modified to hypusine and the corresponding lysine residue in EF-P from at least some bacteria is modified by the addition of a β-lysine moiety. In this review, we provide a summary of recent data that have identified a novel role for the translation factor eIF5A and its hypusine modification in the elongation phase of protein synthesis and more specifically in stimulating the production of proteins containing runs of consecutive proline residues.  相似文献

Using the magnetic particles method, we isolated six polymorphic microsatellite loci from the pea aphid, Acyrthosiphon pisum (Harris), which feeds on a wide range of legume species. The isolated loci were polymorphic, with three to six alleles in 40 aphids. Expected heterozygosities ranged from 0.12 to 0.65. These loci can be used to quantify clonal diversity and compare genetic population structure between sexual and asexual populations.  相似文献

A study was conducted to determine the susceptibility of Anopheles sinensis larvae, malaria vector, from Paju‐shi (Gyonggi‐do) to 24 pesticides including 11 pyrethroids, 11 organophosphates, 1 pyrazole analogue and another pesticide. The mosquito larvae showed the most highly susceptible to chlorfenapyr with LC50 of 0.0063 ppm followed by spinosad, temephos, fenthion and beta‐cyfluthrin with 0.03, 0.0366, 0.0367 and 0.0998 ppm, respectively, and fluvalinate (>128 ppm) was the least susceptible. Also, 11 pesticides of them were compared with Goyang strain (Gyonggi‐do) of data 1992. The development of resistance was noticed to cypermethrin and permethrin as much of 73.59 and 6.23 folds at LC50, respectively, and also fluvalinate was showed high resistance development. In generally, the pyrethroid insecticides were showed higher development of resistance than organophosphorus insecticides.  相似文献
